ABSTRACT Native to subtropical region of South America, yerba mate is responsive to P under some conditions, but the degree of influence of genetic and soil on the growth and composition of the leaf is unknown. The aim of study was to evaluate plant growth, nutrients and potentially toxic elements in leaves of yerba mate clones in response to P application in acid soils. In greenhouse condition, two yerba mate clone seedlings were grown (210 days) in pots, each clone in a completely randomized design in factorial scheme (with and without P; four acid soils). The elemental composition of leaves and the growth of plants were determined. Phosphorus promoted plant growth, but this was not accompanied by increased P in leaf tissue in all conditions tested. The P effect on the elemental composition varied: decrease/null (N, K, Mg, Mn, Cu, Ni, B, Mo, Al, Cd); increase/null (C/N, C, Ca, Fe, V); increase/decrease/null (Zn, Ba, Pb) and; null (Cr). The soils affect the elemental composition of the leaves, especially Mn, with accumulation greater than 1000 mg kg-1. The Ba, Pb, Al and Zn in the leaves varied among clones. Yerba mate response to P was affected by edaphic and plant factors.

ABSTRACT

Early weaning (EW) leads to overweight, visceral obesity, hyperleptinemia, and insulin resistance in adulthood. Treatment with Ilex paraguariensis (yerba mate) improves obesity and insulin resistance in these animals. Here, we evaluated the effects of chronic treatment with yerba mate on the redox balance and liver morphology of overweight early-weaned rats. To induce EW, we wrapped the dams with bandages to interrupt milk access during the last 3 days of lactation. Control pups (C) had free access to maternal milk for the full 21 days of lactation. On postnatal day (PN) 150, EW offspring were subdivided into the EW+YM group, which received the aqueous extract of yerba mate (1 g/kg bw by gavage once a day for 30 days) and the EW group, which received water by gavage for the same period. All rats were euthanized on PN180. The EW group showed higher bound carbonyl (a marker of total protein oxidation), higher TBARS levels (a marker of lipid peroxidation), and lower superoxide dismutase (SOD) activity in liver tissue than the C group, as well as higher triglyceride content and microsteatosis. In plasma, the EW offspring showed higher TBARS levels. One month of yerba mate treatment normalized these parameters. Thus, we have shown evidence that yerba mate improved antioxidant defenses and mitigated liver dysfunction in overweight adult rats that were weaned prematurely.

ABSTRACT

The prevalence of cardiovascular and metabolic diseases is increased in postmenopausal women, which contributes to the burden of illnesses in this period of life. Yerba mate (Ilex paraguariensis) is a native bush from Southern South America. Its leaves are rich in phenolic components, which may have antioxidant, vasodilating, hypocholesterolemic, and hypoglycemic proprieties. This post hoc analysis of the case-control study nested in the Obesity and Bone Fracture Cohort evaluated the consumption of yerba mate and the prevalence of hypertension, dyslipidemia, and coronary diseases in postmenopausal women. Ninety-five postmenopausal women were included in this analysis. A questionnaire was applied to evaluate the risk factors and diagnosis of cardiovascular diseases and consumption of yerba mate infusion. Student's t-test and chi-square test were used to assess significant differences between groups. The group that consumed more than 1 L/day of mate infusion had significantly fewer diagnoses of coronary disease, dyslipidemia, and hypertension (P<0.049, P<0.048, and P<0.016, respectively). Furthermore, the serum levels of glucose were lower in the group with a higher consumption of yerba mate infusion (P<0.013). The serum levels of total cholesterol, LDL-cholesterol, HDL-cholesterol, and triglycerides were similar between the groups. This pragmatic study points out the benefits of yerba mate consumption for the cardiovascular and metabolic systems. The ingestion of more than 1 L/day of mate infusion was associated with fewer self-reported cardiovascular diseases and lower serum levels of glucose. Longitudinal studies are needed to evaluate the association between yerba mate infusion and reduction of cardiovascular diseases in postmenopausal women.

Resumen Introducción: El consumo de yerba mate (YM) (Ilex paraguariensis) es común en Sudamérica y se extiende a todo el mundo. Contiene muchas sustancias bioactivas que pueden ser beneficiosas. También existe la opinión de que puede tener efectos nocivos sobre el ritmo cardíaco, pero no encontramos investigaciones al respecto. Objetivo: Nuestro objetivo fue evaluar el efecto agudo de YM sobre el ritmo, en pacientes enviados para realizar un Holter. Método: Realizamos un estudio clínico seudoexperimental, de tipo antes y después. Tomamos 50 pacientes consumidores habituales de YM y se les realizó un Holter tomando YM y otro luego de 24 sin consumir. También se controló el uso de otros productos que pudieran interferir. Resultados: El 52% fueron mujeres y la edad, de 55 ± 15 años. El 78% tenía al menos un factor de riesgo vascular, el 44% cardiopatía estructural y el 90% tomaba medicación cardiovascular. La frecuencia cardíaca (FC) con y sin YM fue similar. Solo la FC mínima horaria fue menor tomando YM (61 ± 10 vs. 63 ± 9, p = 0.021). No hubo diferencia significativa en la incidencia de arritmias sostenidas, no sostenidas ni en la extrasistolia ventricular. El total de extrasístoles supraventriculares fue mayor sin YM (952 ± 3,538 vs. 1,294 ± 5,201, p = 0.014) y la cantidad máxima horaria también (86 ± 302 vs. 107 ± 360, p = 0.032). No hubo diferencia en la variabilidad de la FC. Conclusiones: En pacientes cardiológicos tomadores de YM, su consumo de la forma habitual no se asoció con cambios importantes de la FC ni con un aumento en la incidencia de arritmias. Hubo menos actividad ectópica supraventricular.


Abstract Introduction: The consumption of yerba mate (YM) (Ilex paraguariensis) is common in South America and is now used all over the world. It contains many bioactive substances that can be beneficial. There is also the opinion that it may have harmful effects on heart rhythm, but no studies have been found on this. Objective: The aim of this study was to evaluate the acute effect of YM on heart rhythm in patients referred for a Holter study. Method: A before and after pseudo-experimental clinical study was conducted by performing a Holter on 50 habitual users of YM, and another one after 24 without YM consumption. The use of other products that could interfere was also controlled. Results: The mean age of the subjects was 55 ± 15 years, and 52% were women. At least one vascular risk factor was found in 78%, with 44% structural heart disease, and 90% taking cardio- vascular medication. The heart rate (HR) with and without YM was similar. Only the minimum hourly HR was lower when taking YM (61 ± 10 vs. 63 ± 9, P = .021). There was no significant diffe- rence in the incidence of sustained or non-sustained arrhythmias, or in ventricular extrasystoles. The total number of supra-ventricular premature beats was higher without YM (952 ± 3,538 vs. 1,294 ± 5,201, P = .014) and also the maximum hourly (86 ± 302 vs. 107 ± 360, P = .032). There was no difference in HR variability. Conclusions: In cardiology patients, usual users of YM, habitual consumption was not associated with significant changes in HR or an increase in the incidence of arrhythmias. There was less ectopic supraventricular activity.

ABSTRACT

Studies have suggested that total energy intake and diet composition affect lifespan and ageing. A high-fat diet induces oxidative stress and affects the development of diseases. In contrast, antioxidants are capable of reducing its harmful effects. Yerba mate beverages are an important source of antioxidants, but there is scarce knowledge about their effects on suppressing fat accumulation. Here, we investigated the compounds present in yerba mate extracts and assessed their effects on Drosophila melanogaster given a high cholesterol diet. LS-ESI-MS analysis showed the presence of matesaponins, phenolic compounds and methylxanthines in all of the examined extracts. In Drosophila, under extract treatment conditions, the mean lifespan was significantly extended from 38 to 43 days, there was an increase in the ability to support induced stress and decrease in lipid peroxidation products. Moreover, yerba mate extracts recovered the glutathione S-transferases (GST) activity and reduced the cholesterol level. Taken together, our results support that extracts can extend lifespan by reducing the detrimental effect of a high-fat diet in D. melanogaster, and this outcome can be associated with the compound content in the extracts. This study improves the understanding of natural interventions that reduce stress-induced oxidative damage, which is fundamental in promoting healthy ageing.

ABSTRACT

DNA repair pathways, cell cycle checkpoints, and redox protection systems are essential factors for securing genomic stability. The aim of the present study was to analyze the effect of Ilex paraguariensis (Ip) infusion and one of its polyphenolic components rutin on cellular and molecular damage induced by ionizing radiation. Ip is a beverage drank by most inhabitants of Argentina, Paraguay, Southern Brazil, and Uruguay. The yeast Saccharomyces cerevisiae (SC7Klys 2-3) was used as the eukaryotic model. Exponentially growing cells were exposed to gamma rays (γ) in the presence or absence of Ip or rutin. The concentrations used simulated those found in the habitual infusion. Surviving fractions, mutation frequency, and DNA double-strand breaks (DSB) were determined after treatments. A significant increase in surviving fractions after gamma irradiation was observed following combined exposure to γ+R, or γ+Ip. Upon these concomitant treatments, mutation and DSB frequency decreased significantly. In the mutant strain deficient in MEC1, a significant increase in γ sensitivity and a low effect of rutin on γ-induced chromosomal fragmentation was observed. Results were interpreted in the framework of a model of interaction between radiation-induced free radicals, DNA repair pathways, and checkpoint controls, where the DNA damage that induced activation of MEC1 nodal point of the network could be modulated by Ip components including rutin. Furthermore, ionizing radiation-induced redox cascades can be interrupted by rutin potential and other protectors contained in Ip.

ABSTRACT

Argentinian native plants Aspidosperma quebracho-blanco, Lantana grisebachii and Ilex paraguariensis are known to have antiinflammatory and antioxidant properties. We demonstrated it in vivo by the redox changes in murine hemolymphatic tissues after infusive extract intake of these plants as revealed in organic trophism, tissue phenolics, hydroperoxides, superoxide, nitrites and γ-glutamyltranspeptidase in thymus, blood and spleen. A. quebracho-blanco reduced hydroperoxidation in blood and spleen of both sexes, with γ-glutamyltranspeptidase negativization in lymphatic organs and thymic nitrosative up-regulation. Males have shown increased phenolic content in blood after treatment. L. grisebachii and I. paraguariensis treatment exhibited incomplete antioxidation and oxidative induction in the studied tissues. Different results according to sex were found in redox response to phenolics and their kinetics, with males showing antioxidant effects, whereas females showed oxidative susceptibility. A. quebracho-blanco exhibited protection of murine tissues against oxidation in both sexes and modulation of their trophism, supporting its therapeutic uses in inflammatory diseases. Also, gender had significant influence in phenolic biodistribution and redox response.

INTRODUCCIÓN: La yerba mate (Ilex paraguariensis) es un vegetal nativo de las regiones subtropicales de Sudamérica utilizada en la preparación de infusiones que se beben en ciertos países de la región. El objetivo del estudio fue determinar los efectos de extractos acuosos de Ilex paraguariensis en los niveles plasmáticos de glucosa, lípidos, urea y creatinina en ratas Wistar. MATERIALES Y MÉTODO: Estudio experimental. Se distribuyeron al azar 20 ratas Wistar en 2 grupos de 10 animales cada uno; el grupo control recibió como bebida agua y el grupo expuesto a yerba recibió como bebida una infusión de Ilex paraguariensis. El periodo de experimentación fue de 60 días, a cuyo término se determinaron las concentraciones plasmáticas de glucosa, colesterol, triglicéridos, urea y creatinina. Las medias de los parámetros fueron comparadas por la prueba U de Mann-Whitney, considerándose significativas las diferencias con p<0,05. RESULTADOS: El consumo de yerba mate disminuyó los niveles de glicemia en el grupo expuesto en comparación con el grupo control, siendo la diferencia altamente significativa (p<0,001). Se observaron niveles significativamente mayores de urea (p=0,04) y creatinina (p=0,02)en el grupo expuesto en comparación con el grupo control. No hubo diferencias significativas entre los niveles de colesterol y triglicéridos entre ambos grupos. DISCUSIÓN: Los resultados sugieren que el consumo de Ilex paraguariensis reduce los niveles plasmáticos de glucosa y podrían poseer un efecto nocivo en la función renal.


INTRODUCTION: yerba mate (Ilex paraguariensis) is a plant species native to subtropical regions of South America and is used to make tea in local countries. The aim of the study was to determine the effects of aqueous extracts of Ilex paraguariensis in plasmatic levels of glucose, lipids, urea and creatinine in Wistar rats. MATERIALS AND METHODS: Experimental study. 20 Wistar rats were randomized in two groups of ten animals each: control group received water while the exposed group received an infusion of Ilex paraguariensis. After 60 days of experiment, plasmatic concentrations of glucose, cholesterol, triglycerides,urea and creatinine were determined. Mann- Whitney U test was used to compare means, differences were considered statistically significant atp <0.05. RESULTS: Blood glucose levels were lower in the exposed group compared with the control group(p <0.001).Plasmatic levels of urea and creatinine were signiticantly higher in the exposed group (p=0.04 and p=0.02 respectively). No significant differences were observed in the levels of cholesterol and triglycerides among the groups. DISCUSSION: The results suggest that consumption of Ilex paraguariensis reduces plasma glucose levels and could have an adverse effect on renal function.

ABSTRACT

O extrato aquoso de erva-mate, obtido a partir de folhas secas de Ilex paraguariensis, é uma bebida amplamente consumida na América do Sul. Inicialmente, nosso objetivo foi caracterizar os compostos presentes nas amostras de erva-mate disponíveis no mercado brasileiro (CH: chimarrão; T: chá mate torrado; G: chá mate torrado, comercialmente acondicionado em garrafas ou C: em copos; TS: chá mate torrado solúvel A mutagenicidade, citotoxicidade e antimutagenicidade de todas as amostras também foram avaliadas atavés do Teste de Ames na presença e na ausência de ativação metabólica. Em seguida, analisamos a amostra TS (2,5, 5,0 e 10 mg/mL) quanto a sua atividade antioxidante e antigenotóxica. Além disso, avaliamos também os efeitos da amostra TS sobre a sinalização da leptina e da insulina no hipotálamo e o estresse oxidativo hepático de ratos adultos obesos programados pela superalimentação neonatal (S). Para induzir S, o tamanho da ninhada foi reduzido a três filhotes por lactante e as ninhadas com número padrão de filhotes (dez/lactante) foram utilizadas como controle. Aos 150 dias de vida, as proles S foram subdivididas em: TS - tratados com extrato aquoso de erva-mate (1g/kg de peso corporal/dia, por gavagem) e S - recebendo água por gavagem durante 30 dias. A prole controle (C) também recebeu água nas mesmas condições do grupo S. Em nossos resultados, verificamos a presença de ácido clorogênico, cafeína e teobromina em todas as amostras analisadas. O conteúdo de compostos fenólicos nas infusões estudadas foram CH: 5,14±0,23; T: 4,33±0,01; G: 0,93±0,25; C: 0,80±0,3 e TS: 8,35±0,5 mg/ml. Não observamos efeito mutagênico ou citotóxico nas amostras analisadas. Um efeito antimutagênico significativo foi observado para a cepa TA97 (pré-, co- e pós-tratamento), na presença de ativação metabólica, em todas as amostras testadas. A amostra TS também apresentou um efeito antimutagênico significativo para a TA102 (pré-, co-e e pós-tratamento)...


Yerba mate extract, made from dried leaves of Ilex paraguariensis, is a tea-like beverage consumed in South America. Here, we aim, firstly, to characterize the compounds present in yerba mate beverages available in the Brazilian market (chimarrao –CH; mate tea –T; mate tea commercially packed in bottles –B and cups ready-to-drink –C; and roasted yerba mate soluble tea –TS). We also evaluated its mutagenic, cytotoxic and antimutagenic properties through Ames test in the presence and absence of metabolic activation. Then, we analyzed exclusively TS sample (2.5, 5.0 and 10 mg/mL) as its antioxidant activity, on plasmid DNA cleavages and upon epithelial esophagus primary culture of Wistar rats treated with a genotoxic agent. Futhermore, we evaluated the effects of mate tea (TS) upon the hypothalamic changes of leptin and insulin signaling and hepatic oxidative stress in postnatal early overfeeding (S) rats. To induce S, litter size was reduced to three pups per dam and normal litters (ten pups/dam) were used as control. In postnatal day (PN) 150, S offspring were subdivided into: S and TS groups treated, respectively, with water or mate tea (1g/kg BW/day, by gavage) during 30 days. C offspring received water. Our results showed the presence of chlorogenic acid, caffeine and theobromine in all analyzed samples. The contents of phenolic compounds in the studied infusions were CH:5,14±0,23; T:4,33±0,01; B:0,93±0,25; C:0,80±0,3 and TS:8,35±0,5 mg/mL. No mutagenic or cytotoxic effect was observed for all analyzed samples. A significant antimutagenic effect was observed for S.typhimurium TA97 (CoT, PreT and PosT), in the presence of metabolic activation, for all tested samples. TS sample also exhibited a significant antimutagenic effect for S.typhimurium TA102 (CoT, PreT and PosT) in the presence of metabolic activation. We did not observed an antimutagenic effect using TA98, TA100 and TA102 for CH, T, C and B samples; and no effect for TA98 and TA100 for TS sample...

Introdução - A dieta hiperlipídica é uma das principais causas da obesidade, provocando importantes alterações metabólicas, como aumento de gordura corporal, dislipidemia e resistência à ação da insulina. Além disso, a obesidade gera um quadro inflamatório crônico e de baixa intensidade no tecido adiposo branco, que é caracterizado pela ativação de vias inflamatórias, como a via do fator de transcrição nuclear kappa B (NF-kB), que é responsável pela transcrição de genes com ação pró-inflamatória, como o fator de necrose tumoral (TNF)-alfa e a proteína quimiotática para monócitos (MCP)-1. A erva-mate (Ilex paraguariensis) contém compostos bioativos, como o ácido clorogênico, a quercetina e o kaempferol, os quais apresentam a capacidade de modular a expressão de genes envolvidos na resposta inflamatória. Objetivo - Investigar o efeito da ingestão do extrato aquoso de erva-mate sobre os parâmetros metabólicos e sobre a resposta inflamatória no tecido adiposo branco de ratos alimentados com ração hiperlipídica. Material e métodos - Ratos machos Wistar foram submetidos à ração controle ou hiperlipídica por 12 semanas. Após esse período, 12 animais de cada grupo foram eutanasiados, constituindo os grupos baseline. O restante dos animais foi distribuído em quatro grupos que receberam, por gavagem, o extrato aquoso de erva-mate (1 g/kg massa corporal/dia) ou água, durante quatro semanas. Após esse período, todos os animais foram eutanasiados. Durante a 12a e a 16a semana do protocolo experimental, os animais foram submetidos ao teste oral de tolerância à glicose e ao teste intraperitoneal de tolerância à insulina. A partir do sangue, foram determinadas as concentrações de glicose, de insulina e de biomarcadores inflamatórios, bem como o perfil lipídico. A composição corporal foi determinada por meio da análise química da carcaça. A partir do tecido adiposo periepididimal, foi avaliada a expressão das proteínas chaves envolvidas na inflamação crônica e na resistência à ação da insulina, por Western blot, bem como a expressão gênica de adipocinas por PCR em tempo real. Resultados - A ração hiperlipídica provocou aumento da adiposidade, alteração do perfil lipídico, intolerância à glicose e inflamação sistêmica. No tecido adiposo periepididimal, a ração hiperlipídica provocou redução da fosforilação da AKT nos animais estimulados agudamente com insulina

ABSTRACT

INTRODUÇÃO: A reação de Maillard ocorre em alimentos termicamente processados e também em sistemas biológicos, em que é chamada de glicação. Esta reação ocorre entre grupos carbonilas e grupamentos aminas e tem especial importância em alimentos, pois promovem alterações sensoriais importantes ao sabor, aroma, aparência e textura. Em sistemas biológicos, entretanto, podem acarretar mudanças em estruturas moleculares que favorecem o estresse oxidativo e participam da patogenia de complicações micro e macro vasculares características da diabetes, aterosclerose e doenças neurodegenativas. Os compostos carbonílicos ou produtos da reação de Maillard (PRMs) formados em alimentos contribuem para o aumento do pool endógeno de compostos carbonílicos. A utilização de substâncias ou alimentos que possam minimizar a formação destes compostos pode constituir-se em uma estratégia para minimizar a sua ingestão. OBJETIVO: Avaliar o efeito da adição de extrato etanólico de erva-mate verde na inibição da reação de Maillard em sistemas modelo alimento. METODOLOGIA: Foram elaborados modelos de biscoito e lácteos com e sem lipídios. Os teores de produtos da reação de Maillard [furosina (FUR), hidroximetilfurfural (HMF), carboximetilisina (CML) e produtos fluorescentes (IF)] foram avaliados. O extrato etanólico foi obtido por extração contínua a quente da erva-mate verde seca e caracterizado quanto aos teores de compostos fenólicos, cafeína e saponinas por cromatografia líquida. A intensidade de fluorescência dos sistemas foi avaliada por espectrofotometria, os teores de FUR e HMF foram analisados por cromatografia líquida de alta eficiência e de CML por ELISA. RESULTADOS: O extrato etanólico de erva-mate apresentou 155µg mg -1 de fenólicos totais, 76µg mg -1 de cafeína, 5µg mg de ácido ursólico e 3µg mg -1 de ácido oleanóico. A formação de HMF, FUR e IF nos sistemas modelo de biscoito não foi influenciada pela adição do extrato de erva-mate.

ABSTRACT

Introdução A obesidade está associada a um quadro de inflamação crônica e de baixa intensidade, que predispõe à resistência à ação da insulina e ao desenvolvimento do diabetes mellitus (DM) tipo 2. O aumento da concentração sanguínea de ácidos graxos não esterificados e de citocinas pró-inflamatórias eleva a expressão hepática de quinases (quinase do inibidor do fator nuclear kappa B (IKK)- e a c-jun N-terminal quinase (JNK)) que aumentam a resposta inflamatória e prejudicam a via de sinalização da insulina. A erva-mate (Ilex paraguariensis) contém compostos bioativos que apresentam a capacidade de reduzir a expressão de genes envolvidos na resposta inflamatória. Objetivo Investigar o efeito da ingestão do extrato aquoso de erva-mate (EAEM) sobre a resposta inflamatória e via de sinalização da insulina no fígado de ratos alimentados com ração hiperlipídica (HL). Métodos Ratos Wistar, machos, foram submetidos à dieta controle (CON) (n= 18) ou HL (n= 18) durante 12 semanas. Após esse período, seis animais de cada grupo foram eutanasiados, enquanto o restante dos animais foi distribuído em grupos que receberam, ou não, por gavagem, o EAEM (1 g/kg massa corporal/dia) durante quatro semanas. Após esse período, todos os animais foram eutanasiados e determinou-se as concentrações de glicose, insulina, colesterol total, HDL-colesterol, LDL-colesterol, VLDL-colesterol, triacilgliceróis (TAG), leptina, adiponectina, TNF-, IL-6, inibidor do ativador do plasminogênio-1 (PAI-1) e a MCP-1 e a atividade sérica das enzimas alanina aminotransferase (ALT) e aspartato aminotransferase (AST). Foi realizada análise histológica para verificação da infiltração de lipídios. Foi avaliada também a composição corporal dos animais. ABSTRACT

PURPOSE: To evaluate the antioxidant effects of enemas containing aqueous extract of Ilex paraguariensis, comparing segments with and without fecal stream and correlating the segments with the duration of intervention. METHODS: Twenty-six Wistar rats were subjected to a diversion of the fecal stream in the left colon by a proximal colostomy and distal mucosal fistula. The rats were distributed randomly into two experimental groups of 13 animals each based on the time of sacrifice after surgical procedure (two or four weeks). Each group was then divided into two experimental subgroups that received either second daily enemas containing 0.9 percent saline solution or aqueous extract of Ilex paraguariensis at 0.2g/100g. Colitis was diagnosed by histopathological analysis and the detection of oxidative tissue damage by measuring the levels of malondialdehyde. The Mann-Whitney test was used to compare the tissue levels of malondialdehyde between colon segments with and without fecal stream in each experimental group, and the Kruskal-Wallis test was used to verify the variance between the levels of oxidative stress according the duration of the irrigation; both tests determined significance at 5 percent (p<0.05). RESULTS: The levels of malondialdehyde in the animals subjected to intervention in the colon with saline with and without fecal stream after two and four weeks of irrigation were 0.05±0.006 and 0.06±0.006, and 0.05± 0.03 and 0.08 ±0.02, respectively. The malondialdehyde levels in the animals irrigated with Ilex paraguariensis with and without fecal stream after two and four weeks of irrigation were 0.010±0.002 and 0.02±0.004, and 0.03±0.007 and 0.04±0.01, respectively. After two and four weeks of intervention, the levels of malondialdehyde were lower in the animals irrigated with Ilex paraguariensis regardless of the time of irrigation (p=0.0001 and p=0.002, respectively). CONCLUSION: The daily rectal application of enemas containing aqueous extract of Ilex paraguariensis decreases oxidative tissue damage in the colon without fecal stream regardless of the time of irrigation.


OBJETIVO: Avaliar os efeitos antioxidantes de clisteres contendo extrato aquoso de Ilex paraguariensis comparando segmentos com e sem trânsito fecal e correlacionando com o tempo de intervenção. MÉTODOS: Vinte e seis ratos Wistar machos foram submetidos à derivação do trânsito no cólon esquerdo pela confecção de colostomia proximal e fístula mucosa distal. Foram divididos de forma randomizada em dois grupos experimentais de 13 animais, de acordo com o sacrifício ser realizado após duas ou quatro semanas do procedimento cirúrgico. Cada grupo foi dividido em dois subgrupos experimentais segundo a aplicação diária ter sido realizada com enemas diários contendo solução fisiológica a 0,9 por cento ou extrato aquoso de Ilex paraguariensis na concentração de 0,2 g/100 ml. O diagnóstico de colite foi estabelecido por análise histológica e os níveis de estresse oxidativo pela dosagem tecidual de malondialdeído Para comparar os níveis de malondialdeído entre os cólons com e sem trânsito fecal em cada grupo experimental adotou-se o teste de Mann-Withney, e o teste de Kruskal-Wallis para verificar a variação dos níveis de estresse oxidativo segundo o tempo de irrigação, estabelecendo-se para ambos o nível de significância de 5 por cento (p<0,05). RESULTADOS: Os níveis de malondialdeído nos animais submetidos à intervenção com soro fisiológico nos cólons providos e desprovidos de trânsito fecal após duas e quatro semanas de irrigação foram de 0,05±0,006, 0,06±0,006 e 0,05±0,03, 0,08±0,02 respectivamente. Os níveis de malondialdeído nos animais irrigados com Ilex paraguariensis, nos cólons providos e desprovidos de trânsito, após duas e quatro semanas de irrigação foram de 0,010±0,002, 0,02±0,004 e 0,03±0,007, 0,04±0,01, respectivamente. Após duas e quatro semanas de intervenção os níveis de malondialdeído foram menores nos animais irrigados com Ilex paraguariensis, independente do tempo de irrigação (p=0,0001 e p=0,002, respectivamente). CONCLUSÃO: A aplicação diária de extrato aquoso de Ilex paraguariensis reduz os níveis de dano oxidativo tecidual nos segmentos cólicos sem trânsito fecal, independente do tempo de irrigação.

Se evaluó la bioaccesibilidad de diversos minerales en infusiones de yerba mate y en preparaciones con leche fortificada con Fe para determinar la influencia de las infusiones en la biodisponibilidad potencial de minerales intrínsecos y del Fe de fortificación. Se evaluó la dializabilidad (D) de hierro, calcio, magnesio y zinc, como indicador de la bioaccesibilidad, en muestras de leche fortificada con sulfato ferroso (SF), bisglicinato férrico (BGF) y EDTANaFe. También se evaluó en mezclas de leche con mate cocido y en infusiones-cocciones con leches fortificadas. En cada caso se estudió la acción del ácido ascórbico (AA) en una relación Fe:AA 1:4. En el mate cocido la DFe fue nula y aumentó a 15% con el agregado de AA. La DFe en leche con SF, BGF y EDTANaFe (1.2; 1.0 y 27.4 respectivamente), se incrementó con el agregado de AA a 2.9; 3.5 y 24.8. La DFe en la mezcla leche:mate cocido resultó 4.7%; 5.7% y 31.2% para SF, BGF y EDTANaFe respectivamente. Con el agregado de AA resultó de 13.5%; 13.7% y 25.7% En infusiones- cocciones en leche fortificada con SF, los valores resultaron menores a los de la mezcla. La DCa no presentó variaciones importantes en las leches fortificadas y sus preparaciones con yerba mate, mientras que la DMg y la DZn solo mostraron una disminución significativa en la infusión-cocción. El consumo de infusiones de yerba mate no aporta cantidades significativas de minerales. Sin embargo, su consumo en mezclas con leche no afectaría negativamente la disponibilidad de Fe en dicha matriz.


The bioaccesibility of various minerals in infusions of yerba mate (mate cocido) and preparations with Fe-fortified milk was assessed to determine the influence of the infusions on the potential bioavailability of intrinsic minerals and fortified Fe. The dialyzability (D) of iron, calcium, magnesium and zinc was evaluated, as indicator of bioaccesibility in milk samples fortified with ferrous sulfate (FS), ferric bisglicinate GF) and EDTANaFe. Dialyzability was also evaluated in mixtures of milk with mate cocido and in infusions cooked with Fe-fortified milk. In each case the effect of ascorbic acid (AA) in a ratio Fe: AA 1:4 was studied. In the mate cocido FeD was zero and increased to 15% with the addition of AA. The FeD in milk with SF, BGF and EDTANaFe (1.2, 1.0 and 27.4 respectively), increased with the addition of AA to 2.9, 3.5 and 24.8. The FeD in the mixture milk: mate cocido was 4.7%, 5.7% and 31.2% for SF, BGF and EDTANaFe respectively. With the addition of AA was 13.5%, 13.7% and 25.7%. In infusions cooked with SF fortified milk, the values were lower than those of the mixture. The CaD did not show significant changes in Fe fortified milk and its preparations with yerba mate, while the MgD and ZnD only showed a significant reduction in infusion cooked with Fe-fortified milk. The use of infusions of yerba mate does not provide significant amounts of minerals. However, its use in mixtures with milk will not adversely affect the availability of Fe in that matrix.

ABSTRACT

Ilex paraguariensis (erva mate) é uma planta amplamente usada na América Latina sob a forma de infusão aquosa. Dentre as propriedades atribuídas a esta planta encontra-se a atividade antioxidante que sugere um papel importante desta droga vegetal na prevenção e tratamento de doenças associadas ao estresse oxidativo como a aterosclerose, fotocarcinogênese e fotoenvelhecimento, entre outras. No entanto alguns compostos presentes nesta planta se encontram sob a forma esterificada, o que poderia dificultar a sua adequada absorção. Uma maneira de aumentar a biodisponibilidade de antioxidantes em extratos vegetais é promover a sua hidrólise visando à liberação dos compostos ativos. O objetivo deste trabalho foi estudar comparativamente o extrato hidroetanólico de Ilex paraguariensis antes e após hidrólise enzimática quanto à composição fitoquímica, atividade antioxidante in vitro e in vivo, biodisponibilidade de compostos antioxidantes e distribuição tecidual destes compostos em animais de experimentação. Para tanto o extrato foi obtido por percolação etanol: água (50 por cento v/v) e sua hidrólise realizada por reação enzimática. A caracterização fitoquímica foi realizada por cromatografia líquida de alta eficiência (CLAE) e espectrofotometria e a atividade antioxidante dos extratos pelos métodos de DPPH e ORAC. Para os ensaios in vivo os extratos (hidrolisado e não hidrolisado) foram administrados oralmente (por gavage) a ratos Wistar machos em sistema de dose única ou doses repetidas (30 dias). Coletou-se o sangue, pele, fígado e cérebro, analisando-se a concentração dos compostos de interesse e a atividade antioxidante destes tecidos pelo método de ORAC. O extrato apresentou boa atividade antioxidante e conteúdo fenólico, sendo que estes valores não foram significativamente alterados pela hidrólise. No entanto, a hidrólise possibilitou uma maior absorção dos compostos de interesse, aumentando a atividade antioxidante plasmática.


Ilex paraguariensis (yerba mate) is a plant broadly used in Latin America as an aqueous infusion. Among its biological properties is the antioxidant activity, which suggests a important role in prevention and treatment of oxidative stress associated diseases, such as atheroclerosis, photocarcinogenesis and photoaging among other. However some of the compounds responsible for that activity are, in crude plant extract, under esterified form, which could make absorption more difficult. An approach to increase the bioavailability of antioxidants from plant extracts is to submit the extract to hydrolysis in order to release the active compounds. The goal of this work was comparatively evaluate the hydroethanolic extract of Ilex paraguariensis before and after enzimatic hydrolysis concerning phytochemical composition, in vivo and in vitro antioxidant activity, bioavailability and tissue distribution of antioxidant compounds in rats. The extract was obtained by percolation with ethanol:water (50 percent v/v) and the hydrolysis performed by enzymatic reaction. The phytochemical characterization was performed by high performance liquid chromatography (HPLC) and spectrophotometry and the antioxidant activity by DPPH and ORAC methods. Hydrolyzed and non hydrolyzed extracts were given orally (by gavage) to male Wistar rats in single and multiple dose (30 days) regimen. Blood, skin, liver and brain were removed, and the concentration of antioxidant compounds and antioxidant activity by ORAC method were evaluated. The crude hydroethanolic extract showed antioxidant activity and phenolic content, but these values were not significantly changed by hydrolysis. However the hydrolysis increased the absorption of the compounds and the plasma antioxidant activity.

ABSTRACT

O objetivo deste trabalho foi avaliar as características químicas e a capacidade antioxidante de folhas de erva-mate (Ilex paraguariensis St. Hil.) originárias da região de Irati-PR (Brasil). Para a caracterização química foram utilizadas as metodologias do Instituto Adolfo Lutz, enquanto a capacidade antioxidante foi determinada pelo método do complexo fosfomolibdênio, utilizando-se ácido ascórbico e rutina como padrões. Nas determinações de umidade, resíduo mineral fixo, lipídios e nitrogênio da erva-mate "in natura" seca foram obtidos, respectivamente, 11,23 %; 5,51 %; 10,84 % e 1,72 %. A capacidade antioxidante do extrato aquoso da erva-mate cancheada foi maior que a apresentada pelo extrato hidroalcoólico, o extrato da erva-mate "in natura" seca apresentou maior atividade. O extrato hidroalcoólico das folhas "in natura" secas apresentou cerca de 50 % da atividade do ácido ascórbico e os demais extratos evidenciaram atividades menores. Com relação à rutina, os resultados obtidos mostraram-se intermediários aos apresentados para os extratos aquoso e hidroalcoólico das folhas "in natura" secas e cancheadas.

OBJECTIVE: The aim of this in vitro study was to evaluate the color stability of a nanofill composite resin (Filtek Z350) in different immersion media. MATERIALS AND METHODS: Twelve resin-based composite specimens were prepared using a silicon cylindrical mold measuring ≅1mm of thickness and ≅ 10mm of diameter. Specimens were light-cured for 40 s from both sides using a LED Radii at 1400 mW/cm² and were randomized into 4 groups (n= 3) according to immersion media: coffee, yerba mate, grape juice or water (control solution). A digital spectrophotometer was used to evaluate the color changes at baseline and at 1-week after immersion in each solution. Specimens were stored in the different staining media for 4 h/day during 1 week. The color differences (DE) were analyzed by paired t-test and one-way ANOVA with complementary Tukey test (p<0.05). RESULTS: After 1-week of immersion, a perceptive color change for the group stored in the grape juice compared to baseline (p=0.008) was observed. Coffee and yerba mate did not show any color changes visible for the human eyes or significantly different from the control group (p>0.05) CONCLUSIONS: The findings of the present study suggest that the tested nanofill resin-based composite was susceptible to staining by substances present in the grape juice.

ABSTRACT

Coffee and yerba maté beverages are widely consumed by the South American population. In adition to the stimulant characteristic, due the presence os caffeine in its composition, these beverages contribute to the intake of bioactive compounds such as phenolic compounds. The content of these compounds justify the several health benefits atributed to yerba maté and coffee: antioxidant, antimutagenic, chemioprevention, antiatherogenic and hepatoprotective effects. The aim of this study was to verify the consumption of these beverages and consequent intake of bioactive compounds (caffeine and total phenolics) by the Brazilian population. A sub analysis of the BRAZOS study (a cross-sectional study) provided data about the beverages intake. The content of bioactive compounds was compiled from original articles and reviews indexed at ISI Web of Knowledge. Coffee (consumed by aproximately 75% of population) and yerba maté (most consumed in the South region) are an important source of bioactive compounds in the Brazilian diet, being chimarrão the major contributor for the intake of these compounds due the large volume ingested.


Las bebidas café y yerba maté son extensamente consumidas por la población de América del Sur. Además de su característica estimulante,debido a la presencia de cafeína en sucomposición, estas bebidas contribuyen para el consumo de compuestos bioactivos tales como los compuestos fenólicos. El contenido de fenólicos justifica los muchos beneficiospara la salud humana atribuidos a estasinfusiones: antioxidantes, anti-mutagénicos, quimiopreventivos, antiaterogénicos y hepatoprotectores. El objetivo de este estudio fue evaluar el consumo de estas bebidas y laconsiguiente ingestión de compuestos bioactivos (cafeína y fenólicos totales) por la población brasileña. Datos sobre el consumo de estas infusiones fueron retirados de un sub-análisisdel estudio BRAZOS (un estudio transversal). Los valores de la concentración de compuestosbioactivos en las infusiones fueron compilados de los artículos originales y revisiones indexadasen la base de datos ISI Web of Knowledge. El café (consumido por aproximadamente 75%de la población) y la yerba mate (consumido más en la región sur) son fuente importante de compuestos bioactivos en la dieta brasileña,siendo el chimarrão el mayor contribuyente para el consumo de estos compuestos debido al gran volumen ingerido.


Infusões de café e erva-mate são largamente consumidos pela população sul-americana. Além da propriedade estimulante, devido à presença de cafeína na composição destes, estas bebidas contribuem para a ingestão de compostos bioativos como os compostos fenólicos. A presença destes compostos biativos estão relacionados a muitos dos efeitos benéficos à saúde humana atribuídos a estes infusos: ação antioxidante, antimutagênica, quimiopreventiva, antiaterogênica e hepatoprotetora. O objetivo deste estudo foi avaliar a ingestão destas bebidas e consequente ingestão de compostos bioativos (cafeína e fenólicos totais) pela população brasileira. Dados sobre o consumo das infusões foram retirados de uma subanálise da estudo BRAZOS (um estudo transversal). Os valores do conteúdo de compostos bioativos nas infusões foram compilados de artigos originais e revisões indexados na base de dados ISI (Web of Knowledge). Café (consumido por aproximadamente 75% da população) eerva-mate (consumida principalmente na região Sul) são importantes fontes de compostos bioativos na dieta brasileira, sendo o chimarrão a infusão responsável pela maior ingestão destes compostos devido ao alto volume ingerido.
